An auxiliary plasmid that expresses a codon-optimized large T antigen (LTag) of simian virus 40 (SV40) in mammalian cells. When co-transfected with a second plasmid that contains SV40 origin of replication (SV40 ori), the LTag will bind SV40 ori and synthesize additional copies of the plasmid in the cell.
Thus, co-transfection with a SV40 ori-containing plasmid will increase the copy number of the plasmid and boost the protein expression from the related plasmid.
The plasmid features the CMV promoter, WPRE stability element, Puromycin resistance gene for selection if needed, and ampicillin resistance gene for bacterial propagation.
