A plasmid for high levels transient or stable protein expression in mammalian cells.
The plasmid features the CMV promoter, WPRE stability element, puromycin resistance gene for selection of stably transfected clones, and ampicillin resistance gene for bacterial propagation.
The plasmid has been used for hundreds of different transfections of numerous protein-expression plasmids; it was successfully used for generating stable cell lines selected for puromycin resistance.
Transient / stable protein expression in mammalian cells
Expression of soluble secreted or intracellular proteins

Boost transient expression of proteins. An auxiliary plasmid that expresses a codon-optimized human RAC-alpha serine/threonine-protein kinase protein (human AKT1; uniprot entry P31749) in mammalian cells. AKT kinases regulate many processes including metabolism, proliferation, cell survival, growth and angiogenesis.
When phAKT1 is co-transfected with a second plasmid into cells, it promotes the proliferation and survival of the transfected cells.
The plasmid features the CMV promoter, WPRE stability element, Neomycin resistance gene for selection if needed, and ampicillin resistance gene for bacterial propagation.
Expression of RAC-alpha serine/threonine-protein kinase 1 (AKT1);
Enhances cell viability and growth during transient protein expression in mammalian cells
Co-transfection with protein-expression plasmids

Boost transient expression of proteins from plasmids that contains the SV40 origin of replication.
An auxiliary plasmid that expresses a codon-optimized large T antigen (LTag) of simian virus 40 (SV40) in mammalian cells. When co-transfected with a second plasmid that contains SV40 origin of replication (SV40 ori), the LTag will bind SV40 ori and synthesize additional copies of the plasmid in the cell.
Thus, co-transfection with a SV40 ori-containing plasmid will increase the copy number of the plasmid and boost the protein expression from the related plasmid.
The plasmid features the CMV promoter, WPRE stability element, Puromycin resistance gene for selection if needed, and ampicillin resistance gene for bacterial propagation.
